Fast Labeling of Technetium-99m-Sestamibi with Microwave Oven Heating

Technetium-99m-methoxy isobutyl isonitrile ("'Tc-MIBI) imaging can be useful to assess myocardial sal11age following t ltrombolysis. In such acute conditions, MIBI must be readi(v a11ailab/e. The standard labeling method requires a 10-min boiling period in a boiling water bath (BWB). The purpose of this study was to compare the BWB technique to a microwa11e o11en heating (MOH) method. Two series of 10 MIBI 11ials (8 cc)-one for BWB and one for MOH-were e11aluated after remo11al of 7-8 cc of head space 110/ume and the addition of /00 mCi (3 cc) of fP"'Tcj-pertechnetate. The two heating processes were e11aluated as follows: (I) BWB: 11ia/s were put in boiling water for /0 min, (2) MOH: 11ia/s were put in a 0.4-cu ft lead-shielded microwa11e o11en for 13 sec. A styrofoam cap protected the aluminum seal from sparking. Labeling efficiency was e11aluated by radiochromatography with Baker flex aluminum oxide plates/absolute ethanol system at I min, 10 min, 6 hr and U hr after reconstitution. The labeling efficiency (percentage of primary complex) was determined and the results showed no significant differences between the two methods. MOH considerably reduces the heating period ( 13 sec) and pro11ides a good labeling efficiency /or in 11i11o imaging.
